Determination of lovastatin from Pleurotus by HPLC-UV method

Abstract

Lovastatin is suitable for lowering cholesterol levels in the body, therefore is used as active ingredient in some cholesterol-lowering drugs. The lovastatin can be introduced in the organism also with different nutrients, like Pleurotus Ostreatus (oyster mushroom), red rice – a fermented rice sort produced using Monascus Purpureus mold.

The main scope of our research is the identification and quantitation of lovastatin in Pleurotus. During our experiments we investigated if the Pleurotus contains the cholesterol-lowering molecule lovastatin and the amount present, using different types of processing of the mushroom.

During our investigations we developed a rapid, precise and reliable analytical method for determination of lovastatin from Pleutorus mushroom using HPLC-UV technique. As result we can say that Pleurotus mushrooms really contain natural lovastatin, the determined amount varies depending on several factors: different processing of the mushroom, sample workup and the origin of the mushroom.

A lovasztatin alkalmas a szervezetben a koleszterinszint csökkentésére, ezért egyes koleszterinszintet csökkentő gyógyszerekben hatóanyagként használják. A lovasztatin emellett különböző táplálékokkal is bevihető a szervezetbe, ilyen például a Pleurotus Ostreatus (laskagomba) a vörös rizs, amely egy erjesztett rizs, amit a Monascus Purpureus penészgomba felhasználásával állítanak elő.

Kutatásunk fő célja a Pleurotusban levő lovasztatin azonostítása és mennyiségi megatározása. A kísérleteink során azt vizsgáltuk, hogy a Pleurotus tartalmazza-e a koleszterinszint csökkentő hatású lovasztatint és ez milyen mennyiségben van jelen a gombában, különbözőképpen feldolgozva azt. Kutatásunk során sikeresen kidolgoztunk egy gyors, pontos és megbízható analitikai módszert a Pleurotusban levő lovasztatin meghatározására HPLC-UV módszerrel. Eredményeként elmondható, hogy a Pleurotus valóban tartalmazza a lovasztatint természetes formáját, a mért mennyiség pedig több tényezőtől függ: különböző elkészítési módok, a minta feldolgozása, valamint a gomba származása.
